Hi, I work with a Gi coupling GPCR protein these days, and we plan to construct a calcium influx assay for GPCR antagonist compounds screening.

As any paper suggests, calcium influx assay is a universal assay for almost all GPCR targets, you just need to build a Gx15/16 overexpression stable cell line, and it should be a great tool for further transit transfect interested GPCR to construct this calcium influx assay.

However, our GPCR calcium influx assay only has a really limited assay window, which is totally not ready for any compound selection.

Then I wish to ask the following questions,

1. Is that true that most GPCR drug targets could follow this method to construct their calcium influx assay?

2. What is the theory of this method?

3. We already did transit transfect both Gx15 and GPCR in ratio 1:1, 1:100, 100:1. but none of them show better results. What should we do next?

4. Did anyone did a similar assay before? Gi protein coupling GPCR in calcium influx assay. That would be really appreciated if anyone could share their protocol.

5. Should we add any Gi-GPCR inhibitor or Knockout Gi to increase the chance of GPCR contact with this Gx15?

6. Where to find a GPCR discussion forum? I really want to learn more cases and experience of GPCR.
