I want to knock a gene using CRISPR/Cas9 technology. Previously I have used lentivirus to knock down gene expression in primary lymphatic endothelial cells (LEC). In primary LEC, I used to use around 10 -20 virus per LEC to knock out gene expression. I am wondering in CRISPR/Cas9 technology, is there any standard virus dosage needed to knock out gene. I will really appreciate if you will suggest me any protocol to follow to optimize virus titer required to knock out gene of interest in LEC or EC. Thank you for your time.
