is there any difference to use protein K and declocker for antigen retrieval during the IHC protocol? what should be preferred?
Yes there is a difference, each antibody you use has to be optimised, both antigen retrieval and concentration of primary antibody etc...
You should consult the spec sheet for the antibody as a starting point.
Most antibodies require heat induced epitope retrieval HIER using a pH buffer usually sodium citrate pH6 or EDTA pH8 or 9.
If you use proteinase k you will need to optimise the digestion time.
