I am working on a project where I need to overexpress VEGF-A isoform 165 aa in NIH 3T3 cells and was planning on using lentivirus with the vector backbone pLVX. I found that the precursor sequence found on NCBI for VEGFA, transcript variant 4 yields an mRNA that will code for a precursor protein that is 191 aa long, and that aa 27-191 from that yields the 165 aa VEGF-A isoform. I'm wondering if it is okay to use this nucleotide sequence that codes for the precursor to achieve the ultimate goal of having overexpression of this 165 aa VEGF in the NIH 3T3 cells?
