I exposed tilapia fish to low, medium and high concentration of malachite green (MG) for 24 hours. What I found was that fish exposed to low concentration are able to accumulate higher concentration of MG in the fish muscle. Whereas fish exposed to medium and high concentration have lower accumulation. This may be due to some mechanism that cause a decrease in permeability of MG into the fish when exposed at a higher concentration. Correct me if I am wrong and please share with me your knowledge for a better understanding of this subject.
Concentrations used, water temperature and pH during experiment should be indicated.
Indeed, when used, malachite green is first quickly resorbed by the gill and cutaneous routes. This resorption is all faster as pH and water temperature are higher. Then malachite green gains blood circulation and is distributed in various tissues and organs. In cells, it is rapidly metabolized, especially in liver. Its metabolites, the main one being leucomalachite green, are distributed in fat, muscles, kidney and liver where they are stored. If removal of malachite green from the body is rapid, that of leucomalachite green is slow, especially in fat. Elimination of malachite green and leucomalachite green is all more rapid as water temperature is higher. Pharmacokinetic studies of malachite green in fish clearly indicate the potential persistence of malachite green and leucomalachite green residues in fish muscle and skin (Afssa, 2002).
If the aquatic is resistant to weak fungi, it slowly and gradually resists the malachite Green's ratio and produces slight effects on the body.
Thank you Prof. Oum Kalthoum for your kind answer. The concentration I used was low (0.05 ppm), medium (0.11 ppm) and high (0.21 ppm). Water temperature was 27 degree Celsius, and pH of the water ranged between 7.2 to 7.4. The experiment for these 3 different concentration was conducted in the same hatchery simultaneously. At this point, I do not think that temperature and pH played an important role because the concentration of MG is considered subtle to change the pH of the water. To add on, MG is a basic dye. Which means, higher concentration of MG should increased the pH of the water if it goes according to the text you mentioned above, and may result in a higher absorbance of MG in the fish muscle. However, my results showed the opposite pattern. Do correct me if I am wrong. Looking forward to your reply.

- how many times have you repeated your experience with these doses, what is the number of replicates that is very important in terms of experimental results?
- in case you have repeated your experiment under the same conditions, have you always found the same results?
- what criteria are you using to choose these doses when it is used as a preventive treatment on fertilized fish eggs at a rate of 2 to 5 mg / L for 20 to 40 minutes, every 2 to 3 days (all days in some farms), throughout the incubation period. and is also applied as a curative treatment to juveniles (fry weighing less than 5 grams) at a rate of 0.5 to 1 mg / L for 20 to 40 minutes, every 2 to 3 days?
