I am currently working on immunoprecipitating a membrane receptor using a monoclonal antibody that has been validated for use in flow and immunohistochemistry. I understand polyclonal antibodies might work better at targeting epitopes (normally hard to target with monoclonal antibodies) in a folded protein. I have not had much success in pulling down this receptor. After WB, coomassie, and silver staining, I can see many bands but after western blot only the HC and LC are visible. Even  LC/MS/MS cannot detect the receptor.Has anybody tried an antibody for an assay other than those for which it has been validated and had success doing so?

Any feedback is greatly appreciated.
