l am trying to HPLC purify a peptide which is rich in polyglutamic acid. Our common running solvents are water and acetonitrile with 0.1% TFA. Is it possible to get rid off TFA after columns and everything else is well conditions after so many years? What kind of running solvents would someone suggest in order to purify a rich glutamic acid peptide? It kind of seems to me it is not so straight forward to change the composition of the mobile phase after so many years.
