In a first part, I started the isolation of strains of actinobacteria from agricultural soil where pesticides are used intensively in order to obtain isolates capable of degrading them but my actinobacteria are very difficult to plant out. The isolation was done in SCA medium, the problem that I had was that my strains did not grow on any medium I tried (ISP1/ISP2/ISP3/ISP4/ISP5/GAUSS/GYEA). My strains do not have the usual appearance of actinobacteria (chalk appearance), are very embedded in the agar and very hard.
It sounds like your initial enrichment lost all the Actinobacter you were looking for. I wasn't sure what SCA medium and the only one I was was for the enrichment of actinomycetes (which are fungi/yeast) so be sure your original enrichment medium is appropriate. But if they aren't growing then you likely don't have them.
Sara Haddad SCA medium might not necessarily support the growth of actinomycetes you desire, especially since it requires a double-layer agar.
Consider the use of other media specifically meant for actinobacteria like AGM, AIM, CPYA, or CPA. You may check their composition in a book: Handbook of Microbiological Media by Ronald M. Atlas
If your strains were obtained from SCA agar. Please try reculture on the same medium again. Once the strain grows well, you may try to transfer the strains to ISP4 or yeast-starch agar.
