I assume the unit used here is square pixels, since the ImageJ-software doesn´t "know" in what magnification you recorded the images. Maybe it is possible to calibrate the images for the wound healing macro by:

- opening an image with a ruler in ImageJ

- drawing a straight line over the ruler (if you hold the "shift"-key the line will be straight)

- then hitting Analyze->Set Scale (a new window opens)

- entering in the "Known distance" the known um of your ruler and in "Unit of length" um (if you check "Global" this will be the scale for all analyzed images you open from this point until you quit ImageJ - to be on the safe site I always write down the values entered).

Usually the "Mean"- and "Min and Max"-values in ImageJ give information about the gray pixel values in the measured picture/selection. "Area" depicts the area of selection in square pixels or in calibrated square units. So "Area" is the proper parameter for you.

We are using image J software for calculating wound areas. We are including a 16cm2 grid in the field. We will select the area of the wound and measure, suppose it is

'x,' then we will take the area of the wound, suppose 'y.' Then once you multiply the x/16 with y you will get the real area. That usually will be a fixed number, not a range.
