You have to check Minimum inhibitory concentration of some isolates and then zone diameter by disc diffusion. Then after plotting you will get a point value which indicates the break point.
Most of the students are confuse for Zone of inhibition and MIC (Minimum inhibitory concentration) . Zone of inhibition means you have to check the inhibitory zone in a specific conc. of active substance using a specific stain. Zone of inhibition express by mm. Disc diffusion method and well diffusion method are the popular methods for determined zone of inhibition. In other scene Mic are the at least minimum conc. drug are active in pathogenic stain. Determine the MIC using the LB medium is the best way I thing.
