We used Sabouraud dextrose agar with chloramphenicol and actidione medium to recover mycelial form of Histoplasma capsulatum at 25 degree centigrade from soil , which was seeded/inoculated with this dimorphic fungus.Further,we isolated yeast phase of H.capsulatum from clinical specimen on Brain Heart Infusion (BHI) agar with blood and cysteine at 37 degree centigrade.The morphology of isolate can be easily studied in Narayan stain,which was developed by me in 1998.Earlier researchers employed mouse inoculation technique to isolate H.capsulatum from soil.You can try Sabouraud dextrose agar with chloramphenicol and actidione from environmental samples by dilution technique.I advise you to inoculate three plates/tubes of media with a solitary specimen and use BOD incubator set at 25 degree centigrade.Further,the inoculated media should be checked daily for 3-4 weeks.The suspected colonies should be examined in Narayan stain to confirm the morphology of H.capsulatum.
