Is there a plasmid vector shorter than pUC19? I'm searching for a very short vector that only has an origin of replication, selectable marker, and promoter region - the MCS/polylinker region and LacZ gene are not required. I need about 2.5-5 nmol of plasmid DNA for my experiments, and with pUC19's length (roughly 2700 base pairs), I would need to generate a lot of bacterial culture to get to that amount (5-10 mg of plasmid DNA). A shorter vector might give me a higher copy number and make the whole process easier.
Update: I will use the plasmid to generate supercoiled DNA (with an intact promoter region) in order to test how sequence-unspecific supercoiled DNA affects the binding between two proteins (at least one of which binds DNA). The binding might require an equimolar ratio between the proteins and the DNA (resulting in the need for such high amounts).

Your rational makes sense to me. Just make sure, that your new plasmid has a high-copy number per cell. Otherwise it won't be of any use.
I am intrigued to know why you need such a huge amount of plasmid DNA with such characteristics?
The pUCmu (1668 bp) and pUCmini (1873 bp) plasmids are available via the BCCM/LMBP Plasmid Collection.
They are based on pUC18, by removing the lacZ fragment and all useless DNA between the active features.

It was constructed by replacing the ampicillin resistance gene in pUCmu by the zeocin resistance gene.
