There are several methods for the separation of fat-related alkaloids in the bottom ethyl alcohol of fenugreek seeds in the extraction device, but is there a simple method and cheap price to get the status of pure alkaloids
I would try one of two methods. Dissolve your methanol extract and partition it between dichloromethane (DCM) and acidic water (I like to use formic or acetic acid since these are easily removed). Remove the aqueous layer, and make it basic with ammonium hydroxide. Extract again with DCM. The free-base alkaloids will be in the DCM layer and can be evaluated with HPLC. This is an inexpensive method.
The other method used strong cation exchange (SCX) columns as per the links below. The column may allow resolution of the alkaloids into different groups.
