           Thanks for your answer. I want to know the actual mechanism of vacuum infiltration which increases the transformation efficiency.

Plant tissue has a lot small air spaces inside and normally even water cannot get in. Vacuum generates a negative atmospheric pressure that causes the air spaces between the cells in the plant tissue to decrease.  When vacuum released, an increase in the pressure allows the infiltration medium, including the infective transformation vector (most commonly in Agrobacterium) to relocate and infiltrated thoroughly into the plant tissue (the small air spaces), as a result,  increasing the agrobacterium contacted tissue or cells and then increasing the transformation efficiency. In plant transformation, some cases vacuum can be applied to a plant part to generate a chimera plant in the presence of Agrobacterium to generate a chimera plant. In that case, vacuum applied period is variable and critical with different plant part, prolonged exposure causes hyperhydricity.
