I am facing problem in wheat regeneration. I got multiple shooting in a regeneration media in wheat during last september, 2014, but currently I am facing no regeneration in same media composition, in same lab, at same temp. I am totally unable to understand the reason behind it.
Plz help me to solve this problem......
Look for the seasonal variations ie in which season you got better results
1. Was the wheat materials from the same genotype? Different genotype may prefer different regeneration medium receipt.
2. Did you use the same stage ('age') materials for the same experiment?
3. Did your chemicals used were the same ones you used in 2014? From different companies? Anything 'expired'?

I am using same genotype which i have used earlier.
and i am using calli of same stage this time also as i used previously.
your third option may be right, because in sept 2014, I was using different lot of MS media and now using different. Although I have ordered new pack of MS media.....may be I will get regeneration this time.....
But I think Mr. Srinath Rao may be correct.......some other people also says same thing...what is your opinion ? .....

You mentioned that you have used different lots of MS powders:
Be aware that there are many mix form of MS powders are available in the market. Some with only MS macronutrients, some with MS macro + micro elements, some also with MS vitamins, and some without adding vitamins in the powder (in this case, you need to buy vitamins to go with it). There are also different vitamins, such as MS original vitamins, and B5 Gamborg's vitamin. So, you need to double-check what you need and what already are in the MS powder you purchased and what are not.
