I have been conducting an experiment which requires to isolate B lymphocytes from human whole blood. Several pieces of literature provide the procedure which includes re-suspension of lymphocytes with buffer after centrifugation. Could someone tell me the reasons for repeating this process a few times?

The pellet is still soaked with soluble components of the blood. Sometimes these can get in the way of subsequent experiments. The repeated centifugations are to dilute them out exponentially.

In addition to John's point another reason could be removal of any residual platelet contamination from platelet rich plasma above Ficoll layer during standard PBMC prep. These will not pellet much at low speeds
