NADPH is generated with ribose 5-phosphate in the pentose phosphate pathway. NADPH has a multi-functional molecule; to help the synthesis of fatty acids and cholesterol, to synthesize reduced glutathione (GSH), to maintain the activity of CYP450, to help synthesize nitric oxide (NO) etc. In the case of neutrophils and monocytes, NADPH is essential for the synthesis of GSH to reduce the redox stress.
