Do you have an idea which compound(s) you would wish to study? The polarity of the compound tends to guide the extraction.
If the compound is unknown, extract with methanol or ethanol. Some people do sequential extraction with hexane followed by ethyl acetate, followed by dichloromethane, then methanol, and finally water.
I agree with above answer you should follow sequential extraction procedure, another way if you want to get best crude extract may follow cold maceration, ultrasonic bath, re fluxing, shaker, dry leaf powder with particular solvent for overnight. Crude extract quality vary with particular solvent.
You should follow successive solvent extraction with solvents in the increasing order of polarity. Soxhlet apparatus can be used. Start with Hexane/Petroleum ether followed by solvents of increasing polarity like ethyl acetate, chloroform, acetone, ethanol, methanol and water. This will help you to fractionate the phytoconstituents effectively and these fractions can be used for isolation as well as in vitro and in vivo studies.
I agree with Jack's suggestion. You can do solvent-solvent partitioning, partitioned with different solvents in increasing order of polarity using a separating funnel, after the methanol/ethanol extraction.
