I am using a three elctrode cell for amperometric detection of glucose. I have modified the working electrode for this purpose. I want to know how much "applied potential" for amperometric detection of glucose vs Ag/AgCl (sat KCl) ref electrode. There is a confusion appears as some of the manuscript cliams -0.56V vs SCE, some claimed +0.40 v VS SCE. Which one is right?
This not a contradiction. The optimum potential depends on the type of electrode and applied modification. You have to find it for your own electrode. Take a cyclic voltammogram. The optimum potential for amperometry is around the potential of oxidation peak (normally 30-40 milivolts more positive than the oxidation peak potential).
You have to experimentally optimize your potential. First take a CV from your electode in the present of glucose, then choose three( or more ) potential containing Ep, a potential after Ep and a potential before Ep . After that use each potential separately in chronoamperometry in the present of glucose, each potential that give you more current, is your opt. pot.
