It depends... Did You try anything yet? I strongly suggest trying CTAB buffer isolation protocol - usually works for fungi, even more 'complicated' ones, for example with ones having strengthen cell wall. It is also the cheapest method. Time needed: app. 4 hours.
Thanks, I didn't Magdalena. as I am trying to find the most used and recommended method. Many articles recommend CTAB. but sometimes Don't find the whole protocol. So I'm afraid to face problem when comes to practical work. also I prefer to use rapid protocol. so i can increase No. of samples in my limited time.
In CTAB You can do as much samples as Your tissue lyser can take at once/twice - in my case about 24 samples in 4 hours. If You are doing it manually - take a day for preparation and than - protocol. If You like - I could send You some checked protocols.
