I bought a new bacterial cellulose strain from ATCC, but I am not quite sure which method is the most effective to store this kind of bacteria for a long term?
We preserve all our bacterial cultures in 20% glycerol broth at -80°C. You can order the glycerol broth from oxoid or prepare them yourselfe. Take from the pure culture on a solid medium enough to make a turbid suspension in the glycerol broth(app. 1 ml in a cryovial) Freese immediately at -80°C. To reculture scrape some material from the frozen vial(don't let the vial thaw) and culture in or at the medium of choice. This way you always use the same culture for all your experiments.
Has anybody got seed lot culture procedure for this? I would like to create a procedure for all of my ATTC cultures. I also designed procedure but i want to be sure that im doing right. If possible please share with me the procedure of this my mail is [email protected] thanks.
