Over the years in our lab, we have used different methods to determine the total protein concentration in dry biomass of different bacteria. Mostly the widely-used Lowry and Biuret methods with bovine serum albumin (BSA) as a standard. We have also used tryptophan-based quantification and calculated the total protein amount based on amino acid analysis of biomass.

However, all these methods have their own pitfalls and, hence, none of the methods produce enough accurate and precise results.
