Could you please provide information regarding the upper limit of RNA sample concentration that the Pico 200 Picodrop Spectrophotometer is capable of measuring? Specifically, I am interested in knowing the highest values achievable for the 260/280 and 260/230 ratios when quantifying RNA samples in terms of (ng/μl). The recorded RNA concentration is noted as 2180 ng/μl. Additionally, the spectrophotometer yielded values of 2.01 for the 260/280 ratio and 1.9 for the 260/230 ratio. I am seeking confirmation as to whether these values have been corrected or adjusted in any manner.
Check the user manual for the equipment. There will be upper & lower limits for how much RNA can be detected.
The easy fix is to dilute the RNA 1:10, measure the concentration of the diluted sample, and then calculate the concentration of the original.
