I know some protocols, but I need the most suitable protocol that might be suitable for Nepal where limited chemicals and equipments are available and without kit (or with kit but should be cheaper than other method).
This is a tricky question. It really depends on the available equipment. If there are no centrifuges at all, then you would need one of those fancy (and expensive) Qiagen kits. It also depends on how pure you need the plasmid.
Nonetheless, I would recommend the alkaline lysis procedure. The procedure is very robust and can be still work even if you make a few mistakes. The chemicals needed are relatively inexpensive and standard (lye, acetic acid, salt, phosphate buffer, ethanol, detergent, etc.). You just need a centrifuge or something to do the spins. I have heard of some researchers constructing hand-cranked centrifuges that worked fine, but I don't have the plans for constructing them.
