I am conducting a thesis study about the antiangiogenic of a fruit extract to duck embryos using CAM Assay, however, I could not find references about the methods in introducing the fruit extract to the CAM. Do you have any suggestions on which method (using a filter paper disc or through direct injection) can the test compounds diffused gradually?
Also, I can't use methylcellulose discs.
By filtering, we prevent to pass some solid unsolved material in some cases. Of course, it doesn't need some cases. For example, for germination of pollen in artificial media contains sucrose, agar and water, there is no difference between the filtering and direct injection.
you can use filter paper soaked in your extract (known concentration) a day before the planting it on the CAM surface. Similarly one can also use gelatin sponge (ribbati et al)
