Timotheus and et al see the multicolor flow cytometry is the only reliable method to purify and characterize ILCs . This is due to the presence of three groups namely ILC1, ILC2, and ILC3, based on the cytokines that they produce. also , They comprise less than 1% of lymphocytes in mucosal tissues and express no unique cell surface markers. Therefore, they can only be identified by combinations of multiple cell surface markers and further characterized by cytokine production in vitro.
you can check my answer to other question. Of course that depends on how many colors you have in the flow cytometry, we have a 10 color flow, so I am using more markers to stain ILCs
STEMCELL Technologies has a kit that can pre-enrich for ILC2s from mouse lung tissue by immunomagnetic negative selection. The protocol only takes 16 minutes once you have a single cell suspension. In naive mouse lung samples, you usually get a 7-fold enrichment. Pre-enriching significantly reduces the time you need on a sorter so it's definitely worth considering. It was used in the Immunity publication attached.
It should work for other tissues as well (ie. gut or tonsil) but might need some optimization.
