I'm looking for an efficient method for isolation of cardiomyocytes without using specific kits
The classical method involves incubating tissue chunks in trypsin at 4 degrees overnight, meaning less hands on time. A more modern methods involves papain and thermolysin digestion for 30 min at 37 degrees. In my experience you will need at least 8 pups per isolation. Both types are protocols should be easily found online.
Hi Semih, I used a solution which was Collagenase and Pancreatin, in a buffer. You need a very high number of neonatal hearts to get a good number of cardiomyocytes.
I can attach the protocol if you think it could work for you.

We also used a solution with Collagenase and Pancreation for the isolation of neonatal cardiomyocytes.
