I want to do TUNEL staining on murine colonic tissue sections which have been paraffin embedded. When doing this so far, TUNEL staining worked great after deparaffinization using xylenes and rehydration using descending concentrations of ethanol. Slides were then permeabalized using 0.1% Triton-X and stained according to the manufacturer's protocol (ROCHE). However, we did not do any antigen retrieval due to this causing high background with the TUNEL staining in the past. Is decloaking/antigen retrieval necessary for counterstaining with Phalloidin or ZO-1? Those are the counterstains we have used so far, with nothing but unspecific binding.
