A very basic query regarding the solubility, stability and storage of carbapenems antibiotics such as Meropenem trihydrate or Imipenem monohydrate. I have known to dissolve them in DMSO in higher concentration for short storage periods but can we use water for the same? Does water affect the stability due to storage in the long run at -20 degree Celsius?
Thanks a lot Saeideh for the help. I am trying to do some kinetics using carbapenems in DMSO solvent. I have observed viscosity problem with DMSO solvent leading to formation of bubbles which interfere with optical density measurement.
Hi Mr. Singh, You can try different percent mixture of water :DMSO , then see which one is good for your study.
Carbapenems belongs to the group of β-Lactam antibiotics. Selection of solvent was based on solubility and stability of antibiotics in solvent system as well as extraction of antibiotics from its formulation. For example, Meropenem is soluble in water as well as chloroform while Imipenem is soluble in methanol and ethanol.
We used Phosphate Buffer to dissolve it for making dilution. It was not dissolved well. Any idea?

I have resolved the issue. Some antibiotics are soluble in ethanol while other require water, especially sodium salts or potassium salts. In your case, I guess you need to dissolve the antibiotics in water, add few drops of conc. NaOH soln (2-4 microlitres) for solubility. Check the MSDS or manufacturer instructions for better ideas.
Imipenem is not very stable, so we only use it fresh and never make aliquots. Meropenem on the other hand is much more stable. We use phophate buffer to dissolve imipenem, and water for meropenem. I suppose the solvent might also be important for the downstream application, and in our case we use the antibiotics for susceptibility testing. One word of advice, never prepare imipenem plates and use them the following day.
If any researcher already did this test then accurate results and suggestion, but if this small test you do in your laboratory by dissolving the antibiotics and also keeping it in different storage place and observed its stability.
Its just my suggestion, suddenly I think and so I am saying this, if it easy to do in small amount , then it will be okay.
