Yes you can use absolute methanol also. But 90% methanol is good for most of the purposes. Normally for efficient extraction, at least 72 cycles are required as a normal rule.You can check for the process completion be taking the cycling solvent (1-2 ml) on a watchglass and allowing it to evaporate. If you see some deposits on the glass, it means still the extraction is required and vice versa in case glass is clear.
The quantity of the methanol is to be taken depends upon the amount of sample. Secondly the different percentages of methanol will extract the things in different proportion. It is also depends upon that which sample you are extracting. Sometimes, you have to take the sample and solvents based on the size of the Soxhlet apparatus you are using.
Choice of solvents / extractants for soxhlet extraction of plant materiall depends on the the nature of plant and chemical compounds it contain. However for gross extraction 80% methanol can be used.
Very first thing should be to remove the chlorophyll. So you should treat the leaves with activated charcoal instead of the extract. That would be more fruitful and easy too.
You have to boil the leaves with the charcoal for sometime. I hope you have done that. Cold treatment would not help too much.
