As you know, for screening mineral-solubilizing bacteria, we use specific mediums such as phosphate (Sperber and Pikovskaya medium) and potassium (Alexandrov medium), etc.

What is the best method to transfer bacteria to these mediums: streaking (from solid medium to solid medium) or inoculating (from liquid medium to solid medium-spot inoculation)?

*When I say "streaking," I am referring to creating a small circle with a loop.

*It is crystal clear that our bacterial isolates must be fresh.

Please share your experiences.
