As you know, for screening mineral-solubilizing bacteria, we use specific mediums such as phosphate (Sperber and Pikovskaya medium) and potassium (Alexandrov medium), etc.
What is the best method to transfer bacteria to these mediums: streaking (from solid medium to solid medium) or inoculating (from liquid medium to solid medium-spot inoculation)?
*When I say "streaking," I am referring to creating a small circle with a loop.
*It is crystal clear that our bacterial isolates must be fresh.
Please share your experiences.
Although it completely depends on your study that either streaking or inoculation which should be preferred but as per ease and also to reduce the chances of contamination streaking your culture (point inoculation) from pure culture plates to the specific medium plates would be the best method also ease of handling varies from person to person so please consider the technique you are comfortable with. Yes, according to me point inoculating your culture from pure culture plate to any specific medium plate with the help of inoculating loop would be easy as well as less resource consuming also. As in point inoculation you can inoculate more than one bacteria over a single specific medium plate by drawing any imaginary partition on plate. Therefore, in this case just by using single plate of specific medium you can test the efficiency or any other targeted property of about 2 or 4 or more number of cultures at the same time.
