Selective Strep Agar is a selective media for all Streptococcus species including Streptococcus pyogenes.The culture plate should incubate anaerobically to form hemolytic zone by bacteria.

Based on the experience of working with various bacteria isolates and testing the activity of various antimicrobial agents against bacteria isolates, I recommend the use of MHB (Mueller Hinton Broth-Oxoid) in your micro-dilutions. This media is known to support the growth of bacteria as well as its constituents do not react with antimicrobial drugs.

You can serial dilute out your antimicrobial drugs (to be tested) using the broth and inoculate standardized bacterial cell suspension. After incubation at 37 degrees Celsius for 18-24 hours, you can subculture by pour plating all wells showing no turbidity (growth) using Trytone Soy' Agar and incubate to determine MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration). 5% sheep's blood agar plates grow S. pyogenes very well. Best growth occurs anaerobically, but the plates can also be incubated aerobically.

I suggest that you should use the 5% Sheep's Blood Agar (SBA) to culture S. pyogenes. This should be incubated at 35°C (anaerobic or ambient air condition) for 24 hours.
