Hi experts,

What is the best storage procedure for mouse gastrointestinal tract samples which will be used for IHC purposes? I had plans to do this experiment before COVID-19 shutdown our lab, but right now I am not even sure my samples are OK. I was about to do a Swiss-roll to freeze them, however, I left them in a 4 degC refrigerator from March till July.

1. Fixed them in 4% paraformaldehyde in PBS.

2. Wash them about 3-5X with PBS and NaAzide.

3. Stored them in PBS with NaAzide for about 4 months in 4 degC.

You may ask why didn't I freeze them immediately? I also thought about that but I was worried the integrity of the tissues may not achieve the best form until I do the Swiss roll. I did not know how long our labs are going to be on shutdown. So yes, I left them in the fridge for 4 months. What is the best thing to do now to save my samples for IHC? Any remedies or is this still OK?

I haven't decided yet what targets to choose, but I wish to do some preliminary investigation between gut-brain-microbiota study. Maybe some immune response related proteins or gut epithelial integrity. At this point, I am not sure yet.
