Hello!

I have been working with 4% PFA solution to fix cells (10 minutes at room temperature) but I noticed that this method disrupts the membrane a lot. I confirmed this by staining the fixed cells with trypan blue to see if the impermeable dye penetrates through the membranes, which it did. My goal is to be able to fix cells without altering the membrane integrity and having similar membrane permeability as live cells. Does anyone have any suggestions on what fixation methods to use for this purpose?
