I'm working with intestinal organoids (colonoids) and I wonder what is the best approach to measure intestinal leakage. I want to measure inside of the matrigel, without transferring them to another plate etc...
What do you think about FITC-dextran (4kDa) together with tight junction proteins expression?
Again I want to avoid removing the organoids from matrigel to avoid disturbing their spatial context.

with FITC, it is possible, in principle:

But, to achieve a bit more physiological scenario, I would rather do micro-injection to load a lumen (unless your organoids are outside-inside) or will transform them into 2D monolayer (where you can also track TEER).
Thanks, Alibek. Indeed TEER can give a more physiological meaning to the leakage process occurring in IBD-derived hIO but 2D monolayer loses the spatial context that 3D organoids have. As the outcome is simply to know if the organoids are leaky I will go for the FITC-dextran protocol. It is also easy and can be used in a high-throughput system.
