A loopful of the plant extract will streak onto a sterile Nutrient agar (for bacterial contamination) and Sabouraud’s agar plates (for fungal contamination). These media plates will be incubated at room temperature for 48h, following which they will be examined for any bacterial or fungal contaminations respectively.
You must take a sample from the plant extract and added it to Petri dishes containing suitable media for bacteria or fungi under laminar air flow conditions , and then incubate the plates under 25 - 30 C in incubator for 2 - 3 days. Then examine the plates , if you see no bacterial or fungal growth , that indicates the plant extract is not contaminated .
Hi, Please take your extract serially dilute using sterile water and spread it on PDA or NA. If there is any emergence of bacteria or fungus indicates your extract is contaminated.
I prefer to pass the whole plant extracts through a millipore 0.22 micro filters, so you will have sterilized plant extract from any fungal contaminants . If you want to check entierly ,you can use PDA medium after using millipore filter>
If you want to check if your extract is contaminated by fungi and be sure that it is not a bacterial contamination, spread it on medium for fungi (PDA or Saboraud's agar) supplemented with antobiotic (to avoid bacterial growth) such as chloramphenicol, ampycillin or oxytetracycyline.
Really i agreed with dear Oadi Manty ,First you can filter extract with either 0.45 or 0.22 micron millipore , then if want confirmation, you can check with PDA or any medium for Fungi
