In synthesis process of curcumin loaded albumin nanopaticles, when I used EDC as crosslinker, surprisingly it caused intense aggregation! Actually I used these methods :

Does anyone have any experience what the problem is? Could the problem be the ratio of albumin to EDC? In this case, what is the best ratio?

Instead of the amount of EDC, I would suggest break the reaction into 2 step, using EDC and NHS。. This will help prevent aggregation between the protein itself
I don't think it's surprising! As you have a protein with multiple amine and carboxyl groups, you simply have too much EDC and are causing excessive crosslinking. There is no 'best' amount of EDC that can be applied in all applications. The 'correct' amount will be determined by concentration of the BSA and other reactants, buffer type, pH, temperature, time of reaction, quality of the EDC etc. I think you just need to titrate the amount of EDC, and clearly the optimum amount will be below the level you used last time.
