I have a plasmid that has a N-terminal FLAG-tagged protein preceded by a pre-protrypsin leader. The WT protein has a GPI anchor, that presumably is intact here.

I looked on Milipore, and it says "The preprotrypsin leader sequence3 precedes the FLAG sequence and directs secretion of the fusion protein into the culture medium".

It is my understanding that my plasmid is supposed to express my GPI-protein of interest to the cell membrane. Is this not true because of the pre-protrypsin signal?
