It depends on the microbes being tested. Disc diffusion is not applicable to all microbes. You can refer to the previous discussion.

You start with basic techniques (agar diffusion using discs or wells,...) to know the antimicrobial effect (presence or absence of antimicrobial activity), then you select only the bioactive sample to be tested for MIC calculation. It's not logic to realize the reverse protocol.
