For my 2 samples i am finding Polyphenol concentratiOn.
For sample one: O.D of different concentration (high to low) are 1.777, 1.209, 0.915, 0.761. I select 0.761 for Y value and find 0.26 g/L polyphenol.
For sample 2nd: O.D of different concentration (from high to low) is 2.402, 1.720, 2.022, 1.170, 0.944. I select value of Y 0.944. And find polyphenol concentration 0.31 g/L.
Is the Y value for 2nd sample is correct?? And which sample is good one in polyphenol concentration??
As you might known, the simplest method for the determination of total polyphenol compound from the samples is calculate based on a standard curve of gallic acid or whatever (phenolic-compound, depend on your sample-origin). The important thing is your samples' OD measurement should be within OD value ranging of the standard as you prepared and measured (if the values are too high or very low, you have to dilute or concentrate the samples), and normally, OD value lower is more accurate. Hope it useful for you!
Our practice has shown that in conducting an experiment to establish a standard curve with gallic acid, the linear range for a correct measurement is in the range of less than one. If you dilute the concentration of polyphenol you had (0.26 g / l) in a 1: 1 ratio you should get good results ... I wish success!
Nikolay Solakov thanm you for your response . You mean to say that i futher dilute my sample 2.
And if i have a many value less then one then which value will be good for Y the value that is more diluted one??
Cuong Van Tran why we select y value less than 1 and do have any reference. Because i have to explain this in my presentation with reasons.
The answer to this question can be seen in the results of a properly conducted experiment to construct the calibration curve with gallic acid. there is a rectilinear stretch within the range of concentrations that provide absorption with a value less than one, which means that working with such concentrations will have accurate results. Success of the presentation!
