In my opinion, the most suitable quantification method depends on what compounds you are interested in. Plants have several classes of compounds with phytoterapeutical properties. For example, important classes are polyphenols, flavones, tannins, volatile oils, polysaccharides etc. In this classes, many chemical compounds have antioxidant properties (eg. gallic acid, rutin, quercetin, linalool…)
You can express total polyphenols/flavones/other classes using spectrophotometric methods (most convenient). One of this methods is Folin-Ciocalteu for total polyphenols. For volatile oils, GC methods are very popular, while specific polyphenols, flavones, tannins, polysaccharides could be easily determined using HPLC or TLC/HP-TLC.
Find attached some research articles that could be useful. Sometimes, several extractions in different polarities solvents should be done as preliminary treatment and then fractions analyzed separately.
Phytochemical screening includes quantification of all plant based bioactive molecules: phenolics, flavonoids, tannins, saponins, anthrocynidins. For antioxidants first get total phenolic and flavonoid content, quantify the single molecules using UPLC/HPLC and for bioactivity: ABTS/DPPH invitro assays or H2O2 cell based assay.
