i am gonna to link two secondary OH groups of cyclodextrin and carboxylic groups of histidine and arginin. i have tried many coupling reagent such as TEA-CDI which first OH converts to amine then we can bind them by EDC NHS. finnaly i decided to use estrification but i dont know EDC is better than DCC or not? if yes please cheak my title question.

1. EDC might be easier to remove, use some 10% molar excess.
2. Catalytic amount (5-10 mol%) of DMAP might be enough.

4. It may worth an attempt to try transesterification. E.g. using p-nitrophenyl ester. A similar reaction is described in DOI: 10.1007/BF00660746 for alpha CD but the beta is also a very good host for p-nitrophenyl.

Thank you for your replying but i have to say i used HOBt and Methylmorpholin and i think we dont have any catalyst like them, they are the best to amino acid binding.@laszlo jicsinszky
