I have decidet to bought Rhodamine 123 to evaluate mitochondrial activity of a treated green algae. In the research (published) paper where the protocol is described, it is written that I have to dissolve R-123 in 0.1 M PBS.
I would like to know what does exactly means 0.1 M in the case of PBS which is a mix of different salts.
At the moment I have PBS 1X for in vitro human cell culture from Invitrogen and Lonza and PBS tablets from Sigma. But I'm not sure they can be used!

Just make a 10x PBS and you will have your required concentration of solution.
800 mL ddH20 in a larger beaker with a stirbar then add:
80g NaCl
2g KCl
7.62g Na2HPO4
0.77g KH2PO4
pH to 7.4
QS to 1L with ddH20
Re- check the solution to make sure it is still pH 7.4
This should give you 0.1M PBS
