I searched several buffer recipes for flow cytometry, and I need to include DNase to reduce DNA-associated cell clumpiness. Some protocols mention DNAse II while others mention DNAse I. Any ideas on what I should use. I am working with human cancer cells. Thanks!
I'm using DNAse I to prepare cell suspension with low level of cell aggregates from murine and human cardiac tissue, murine breast cancer, LLC and EL-4 carcinomas. Cell aggregates is always less than 5% (both SSC/FSC and counting under the microscope).
I use DNase I (150 ug/ml) from Worthington Biochemical to reduce cell clumping during skeletal muscle tissue digestion. I've found that sometimes my samples will develop clumps long after the digestion step when preparing the samples for flow cytometry, so I've been thinking of adding DNase to my flow buffer. What concentrations are you using for this?
