When imaging immunofluorescent stained tissue slides I am seeing these strange rings of dots. They appear on and off the tissue - just all over the slide. Has anyone seen anything like this before? Is there a way to resolve it?
I haven't seen something similar but i could speculate.
Since this is not a specific staining but an artifact and since there is a specific ring-like pattern, then a probable cause could be air bubbles that formed in these areas during the incubation process and then the slide dried and trapped the antibodies in these areas around the air bubbles.
Again, i am not certain about this but it could explain the non specific pattern.
I have seen similar patterns with certain mounting mediums. Some are prone to form very small air bubbles and to aggregate antibodies like that. I suggest doing more washing before mounting, to eliminate everything that did not bind.
