We're working on energy metabolism of human white blood cells. We measure mitochondrial respiration right after isolation and after 4hours of culture and treat them with various solutions. For all our samples including an untreated control we observe a decline in respiratory activity after 4 hours of culturing in RPMI at 37°C.

Viability doesn't seem to be the problem. Does anyone have an idea what could cause a decline in metabolic activity after 4 hours in vitro?
