I've just joined a project in protein production.
We have an issue when growing E.coli in exactly the same conditions, same media, same cell line, same flask type, same incubator etc, some of the cultures begin to decrease in OD, whereas others increase.
We have ruled out phage too.
Any ideas?
Are the flasks cleaned well and thoroughly rinsed with double-distilled water prior to autoclaving?
Is the pH of the media uniform in each flask?
Does the OD only decrease after adding inducer?
Are all of your E. coli cells at the same growth phase and cell density when inoculating?
Is there an extreme temperature gradient in your incubator? (ie could cultures near one part be exposed to significantly higher temperatures than the rest of the cultures)
OD tends to measure living and dead cell , you should expect to see decreasing in OD at the beginning of your work especially when the environment (media ) don't having osmotic support
