I am new to liposome making. My drug (carbamazepine, mol wt. 236) is highly insoluble in the water. I tried to load it in bilayer using a basic protocol. After lipids(DMPC, DMPG, Cholesterol) mixing with dissolved drug in organic solvent (acetonitrile), and drying, hydration of lipid film with water was not possible. I tried to add a little bit of methanol to dissolve it but, it remained undissolved looking like precipitated chunk. Is there a protocol that you can recommend that I can start with? How should I dissolve the lipid film?

After drying, you need to add PBS/water to the thin layer, and sonicate it above 60C (or above lipid transition temperature) to synthesize drug loaded lipoomes. I did my experiments of lipid shelled bubbles and you can see the publications for further details

Your lipid's transition temprature is important. You must study this degree and add the glass bubles. And evoporation pressure is important for acetonitrile.
