I am working with hepatocellular carcinoma cells. I want to study oxygen consumption in these cells. Is there a kit or a procedure I can use other than the XF Cell Mito stress kit?
Hi Ezel: In terms of measuring oxygen consumption in tumour cells, I would recommend the CCK8 assay, which measures cellular dehydrogenase activity that comes principally from mitochondrial respiration. The dehydrogenases are oxidoreductase enzymes that catalyze the oxidation of a substrate through the transfer of hydrogen (hydride) to an acceptor (or co-enzyme). In the case of the CCK8 assay, the acceptor would be WST-8, which is then transformed into a formazan salt. Since this is an oxidation reaction that occurs in oxidative phosphorylation, I believe it would be a good measure of oxygen consumption.
