Not only for macromolecule: solid NMR is a very interesting analytical method to distinguish between polymorphic forms of API (small molecule crystallized). And one advantage of this technique is that signals of small molecule are far from the signals of excipient allowing survey of polymorphism determination in drug product manufaturing process by doing solid NMR directly on capsule or tablets.
One remark, do you know that the magic angle used in solid NMR technique is the same angle used to build pyramid? strange?isn'it.
solid state NMR is used for large miolecules (proteins,etc ) that have low solubility iin common NMR solvents. I'am not sure what kind of results you'll get though cause i have not used it before.
Solid state NMR is for insoluble macromolecules, especially nanomaterials like MOF and nanoparticles. At this point, solid state NMR has advantages over solution NMR. But the signal solid state NMR showed is very broad in comparison with solution NMR, so the resolution is not as good as solution NMR.
But if we try to take solid state NMR of a compound which is soluble in water or other organic solvents, how will its spectrum be different from spectrum obtained by solution state NMR and what extra information we can get using solid state NMR?
In solid samples, the molecules will not tumble in all directions as they do in liquid samples. This will, as previously stated in this topic, result in much broader signals in solid state NMR. This can be overcome (at least to some extent) by special techniques, such as high resolution magic-angle spinning (HR-MAS), by cross polarization, by using high power pulses and so forth...So, unless dealing with very large systems, one could possibly obtain the same type of information that is available from a corresponding liquid NMR spectra, without losing too much in resolution. For solid samples, the sample preparation is perhaps the most crucial part...as structural disorder or inhomogenity will give lousy spectra.
